DNA fragmentation and membrane integrity in sperm of farmed Arctic charr (Salvelinus alpinus)

چکیده
Despite the relatively long history of captive breeding, Arctic charr still exhibits a generally low, but highly variable reproductive performance in aquaculture. A recent publication exposed potential paternal factors influencing outcome broodstock from Swedish breeding program. Interestingly, factor appeared to be more closely connected embryo survival than fertilisation rates. This lead speculations on whether e.g. chromatin related issues, potentially oxidative stress could involved. In order investigate this hypothesis present study assessed levels DNA fragmentation, using SCD-method, and membrane integrity, flow cytometry, sperm farmed charr. Moreover, existence associations was tested between fragmentation integrity individual semen samples viability their resulting progeny. We found high sires, ranging 24% 86% with median 67%. Membrane values were high, 93.1% 99.6% viable cells, 98.8%. moderately correlated (r = 0.304, p < 0.05). Fertilisation rates proportions eyed eggs showed substantial variation 0.497, However, large differences proportion rate, 52% 81.6% respectively, highlight that main loss occurred due mortality rather failed fertilisation. No correlation either or (fertilisation rate eggs) sires. Overall, our identified very which influence fertility question thereby mitigating mechanism involved low success most often observed Further exploration relationship would needed, though.

Behaviour, physiology and carotenoid pigmentation in Arctic charr Salvelinus alpinus.
The behaviour during an exploration task and the response to a confinement stress of Arctic charr Salvelinus alpinus were evaluated. Behaviour of individuals during 90 min of exploration was classified into high and low activity. High-activity individuals had higher plasma cortisol levels following stress compared to low-activity individuals. This indicates that high- and low-activity individua...
متن کاملThe developmental transcriptome of contrasting Arctic charr ( Salvelinus alpinus) morphs
Species and populations with parallel evolution of specific traits can help illuminate how predictable adaptations and divergence are at the molecular and developmental level. Following the last glacial period, dwarfism and specialized bottom feeding morphology evolved rapidly in several landlocked Arctic charr Salvelinus alpinus populations in Iceland. To study the genetic divergence between...
متن کاملHolarctic phylogeography of Arctic charr (Salvelinus alpinus L.) inferred from mitochondrial DNA sequences.
This study evaluated mitochondrial DNA (mtDNA) sequence variation in a 552-bp fragment of the control region of Arctic charr (Salvelinus alpinus) by analyzing 159 individuals from 83 populations throughout the entire range of the complex. A total of 89 (16.1%) nucleotide positions were polymorphic, and these defined 63 haplotypes. Phylogenetic analyses supported the monophyly of the complex and...
متن کاملThe effect of anesthetics on carotenoid pigmentation and behavior in Arctic charr (Salvelinus alpinus)
Status of individual animals is often shown in pigmentation. For instance, stress responsiveness is associated with melanin-based pigmentation in vertebrates in general. This pattern is evident in Atlantic salmon (Salmo salar) and rainbow trout (Onchorhynchus mykiss) with melanin-based spots indicating stress coping style. Arctic charr (Salvelinus alpinus) differ from other salmonids in pigment...
متن کاملDominance and stress signalling of carotenoid pigmentation in Arctic charr (Salvelinus alpinus): lateralization effects?
Social conflicts are usually solved by agonistic interactions where animals use cues to signal dominance or subordinance. Pigmentation change is a common cue used for signalling. In our study, the involvement of carotenoid-based pigmentation in signalling was investigated in juvenile Arctic charr (Salvelinus alpinus). Size-matched pairs were analysed for pigmentation both before and after being...
